KPV is a short, naturally occurring tripeptide consisting of three amino acids: Lysine (K), Proline (P), and Valine (V). It is the terminal sequence derived from alpha-melanocyte-stimulating hormone (‭$\alpha$‬-MSH), retaining the parent hormone's potent anti-inflammatory properties without triggering melanin production or skin-darkening side effects. It operates through several primary mechanisms: • NF-‭$\kappa$‬B Inhibition: It blocks the activation and nuclear translocation of Nuclear Factor kappa B (NF-‭$\kappa$‬B), a master regulator of inflammatory pathways. This significantly reduces the production of pro-inflammatory cytokines like TNF-‭$\alpha$‬, IL-6, and IL-1$\beta$. • Intracellular Uptake via PepT1: Unlike many peptides that work solely via cell-surface receptors, KPV enters cells through the PepT1 transporter, which is often upregulated at sites of tissue inflammation (such as the gut lining). • Antimicrobial & Barrier Repair: It exhibits inherent antimicrobial activity against pathogens like S. aureus and C. albicans, while supporting mucosal and epithelial tissue recovery. Administered orally, topically, or via subcutaneous injection, KPV is heavily researched and utilized for inflammatory bowel disease (IBD/colitis), autoimmune skin conditions (eczema, psoriasis, rosacea), and as an inflammation-reducing component in tissue recovery protocols.
